The saying is often used to suggest betrayal, especially by someone who claims to have friendship for you. So, you can either have a character say that to another "et tu, Brute?" as a way of saying "and you're going to stab me in the back, too, buddy?" or you can just have the concept: betrayal by someone who claims loyalty.
